P-107: Vitrification and In Vitro Culture of Mouse Preantral Follicles in Presence of Growth Factors

author

Abstract:

Background: Cryopreservation of oocytes is an effective technology in assisted reproductive technology. Although successful vitrification of gametes has been reported in several mammalian species, the survival is generally low. The aim of this study was to investigate the effects of fibroblast growth factor (FGF) and hepatocyte growth factor (HGF) on preantral follicle development after vitrification in vitro. Materials and Methods: Preantral follicles with diameter of 150-180 μm were mechanically isolated from 18- 21 day old NMRI mouse ovaries. Follicles were vitrified and warmed and then cultured in α-minimal essenstial medium supplemented with 0,20 ng/ml FGF,20 ng/ml HGF, 20 ng/ml from each of FGF and EGF. After 12 days hCG/EGF was added to culture medium and after 18- 20 h later the presence of cumulus oocyte complexes (COC) and oocyte maturation state assessed. Results: The results of this study showed that the rate of MII oocytes in FGF group increased in comparison with control and other treatment groups (p<0.05) There was significant increase in survival rate of follicles in FGF-HGF group in comparison with control and other treatment groups (p<0.05 ). After in vitro ovulation induction the follicles in HGF group showed a lower ovulation rate (p<0.05) than those cultured in other groups. Conclusion: FGF in mouse vitrified follicle culture system increases oocyte maturation rate and HGF decreases COCs number. Addition of HGF and FGF simultaneously to the culture medium increase follicle survival.

P-83: Development of Mouse Preantral Follicles in Fibrin-Alginate Matrix during In Vitro Culture

Background This research was conducted to assess preantral follicle development in fibrin alginate matrix following ovarian tissue vitrification MaterialsAndMethods Ovaries of 13-day-old NMRI female mice were removed and placed in control and vitrification groups. Vitrification group ovaries were transferred in media containing ethylene glycol, dimethyl sulphoxide, and sucrose then were plunged...

P-116: Maturation Genes Expression during In Vitro Culture of Vitrified Mouse Preantral Follicles

Background: Evaluation of maturation genes expression and their pattern during in vitro culture of vitrified preantral follicles is an essential and could promote either vitrification procedure or culture condition. Materials and Methods: Preantral follicles (with 100- 130 μm diameter) isolated mechanically from 12-14 days old NMRI mice ovaries and divided into vitrification and control groups....

The Effects of Fibroblast Co-Culture and Activin A on in vitro Growth of Mouse Preantral Follicles

Background: This study was conducted to evaluate fibroblast co-culture and Activin A on in vitro maturation and fertilization of mouse preantral follicles. Methods: The ovaries from 12-14-day-old mice were dissected, and 120-150 μm preantral follicles were cultured individually in α-MEM as based medium for 12 days. A total number of 456 follicles were cultured in four conditions: (i) base mediu...
